Cusco served as the capital of the Inca Empire before becoming a Spanish colonial city built literally on top of it — many colonial church and building foundations are visibly Inca stonework beneath newer construction. Most Machu Picchu itineraries begin here, with a day or two for altitude acclimatization first.
